If you do have an unusable rune or glyph that hasn't yet been applied to your gear, you can move it to a different piece gear by using the upgrade Glyphs context menu in the Glyphs panel. This will create a new Glyph Set with the selected glyph. If you are looking to remove a glyph from an item then you'll need to salvage it with an Black Lion Salvage Kit or Ascended Salvage Tool.
